Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous albumin

  • Recognizes human albumin
  • Albumin most abundant protein in blood plasma
    • Produced by well-differentiated hepatocytes
    • Plays role in maintaining osmotic pressure
    • Decreased levels of serum albumin may be sign of kidney or liver diseases and diabetes

Process:

  • Antibody specific for Albumin pre-coated onto microwells
  • Albumin protein in samples captured by the coated antibody after incubation
  • After extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for Albumin is added to detect captured Albumin
  • For signal development, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
