The BUB1 gene encodes a serine/threonine-protein kinase that is integral to the spindle assembly checkpoint, a crucial mechanism ensuring proper chromosome alignment and segregation during mitosis. BUB1 is involved in detecting and correcting errors in kinetochore-microtubule attachments, which is vital for maintaining genomic stability. It acts by delaying the onset of anaphase until all chromosomes are correctly attached to the spindle apparatus. BUB1 is recruited to kinetochores in early mitosis, where it phosphorylates several substrates to activate other checkpoint proteins, such as BUBR1 and MAD2, thereby preventing premature progression through mitosis.
