Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous human CCL3

  • Recognizes human CCL3
  • Chemokine (C-C motif) ligand 3 (CCL3) also known as MIP-1α belong to family of chemokines
    • CCL3 found in CNS and receptors CCR1 and CCR5 expressed by astrocytes, microglia and neurons
      • CCL3 and receptors contribute to development of bone disease in multiple myeloma by supporting tumor growth and regulating osteoclast differentiation
    • CCL3 associated with regulation of cell growth, angiogensis and metastasis of tumors (melanoma, renal cell carcinoma and colorectal cancer)
    • CCL3 enhances cell migration and metastasis by up-regulating matrix MMP-2 expression in chondrosarcoma cells

Process:

  • Antibody specific for human CCL3 pre-coated onto microwells
  • Human CCL3 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for human CCL3 added to detect captured human CCL3 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
