Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous CHGA

  • Recognizes human CHGA
  • Chromogranin A is a member of granin family of neuroendocrine secretory proteins
    • Precursor to functional peptides (vasostatin, pancreastatin, catestatin, parastatin)
      • Negatively modulate neuroendocrine function of releasing cell (autocrine) or nearby cells (paracrine)
    • CgA one of most used tumor markers in NET's
    • Elevated CgA concentrations in serum or plasma of patients with different types of tumors
    • Abnormal concentration found in non-malignant diseases (renal failure, heart failure, proton pump inhibition, gastritis, hypertension) or patients with liver diseases

Process:

  • Antibody specific for CHGA pre-coated onto microwells
  • CHGA protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for CHGA is added to detect captured CHGA protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
