Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous CNPY2

  • Recognizes human CNPY2
  • CNPY2 (MSAP, TMEM4, ZSIG9) is a positive regulator of neurite outgrowth
    • Interacts with ezrin-radixin-moesin (ERM)-like myosin regulatory light chain-interacting protein (MIR) and two proteins co-localized in cell lines and primary neurons
    • HIF-α regulated, secreted angiogenic growth factor that promotes smooth muscle cells migration, proliferation and tissue revascularization
    • Enhances cell spreading in fibroblasts and migration of rat C6 glioma cells

Process:

  • Antibody specific for CNPY2 pre-coated onto microwells
  • CNPY2 in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for CNPY2 is added to detect captured CNPY2 protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
