Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous ENO2

  • Recognizes human ENO2
  • ENO2 (enolases) are cytoplasmic glycolytic enzymes that may be involved in differentiation
    • Three isoenzymes:
      • Alpha: expressed in most tissues
      • Beta: expressed in muscle tissue
      • Gamma (ENO2): isoform neuron-specific enolase (NSE), a homodimer is localized in neurons and neuroendocrine cells
        • Cancer diagnostic marker for brain tumors
        • Plays role in glycoloysis-related energy pathway
        • Might be involved in higher metabolic activity during day

Process:

  • Antibody specific for ENO2 pre-coated onto microwells
  • ENO2 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for ENO2 is added to detect captured ENO2 protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
