Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous F12

  • Recognizes human F12
  • F12 (Factor XII, FXII, Hageman factor) is a 80 kD single chain glycoprotein circulating in blood as inactive zymogen
    • Plays role in blood coagulation, fibrinolysis, kinin generation
    • F12 bound to surface when plasma comes into contact with negatively charged surfaces (glass, kaolin, dextran sulfate), undergoes limited proteolysis by plasma kallikrein creating two forms
    • F12 inhibition has protective role against arterial thrombosis and stroke suggesting F12 may be a target for antithrombotic therapy

Process:

  • Antibody specific for F12 pre-coated onto microwells
  • F12 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for F12 is added to detect captured F12 protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color developement and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
