- Recognizes human GM-CSF
- GM-CSF (CSF2) is an important hematopoietic growth factor and immune modulator is produced by a variety of cell types (T cells, macrophages, endothelial cells, fibroblasts) upon receiving immune stimuli
- Recognizes as a stimulator for proliferation of granulocytes and macrophages from bone marrow precursor cells
- Shown to promote survival and activation of mature myeloid cells, contributes to maintenance of innate immune homeostasis
- GM-CSF has proinflammatory functions and plays critical roles in development of autoimmune and inflammatory diseases (Th17 driven diseases)
- Role in embryonic development by functioning as embryokine produced by reproductive tract
Process:
- Antibody specific for GM-CSF pre-coated onto microwells
- GM-CSF protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for GM-CSF is added to detect captured GM-CSF protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
