Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous ICAM-1

  • Recognizes human ICAM-1
  • ICAM-1 (Intercelluar adhesion molecule 1, CD54) is a transmembrane glycoprotein of immunoglobulin superfamily
    • Critical for firm attachment and transmigration of leukocytes out of blood vessels and into tissues
    • Expressed by several cell types, typically on endothelial cells and cells of immune system
      • Expression can be up-regulated by various stimuli
    • Ligand for LFA01 and Mac-1 serves as receptor for rhinovirus and one of several receptors used by Plasmodium falciparum
    • Soluble form (sICAM-1) measured in various body fluids, with increased levels being found in serum with patients with cancer, cardiovascular disease, cancer and autoimmune diseases

Process:

  • Antibody specific for ICAM-1 pre-coated onto microwells
  • ICAM-1 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for ICAM-1 is added to detect captured ICAM-1 protein
  • For signal development, Stretavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
