Description

Determine the ability of an organism to split indole from the tryptophan molecule. Pro-Lab Diagnostics™ Kovac's Reagent is a spot indole reagent that is used in the qualitative method of identifying tryptophanase activity in microorganisms, such as Escherichia bacterium.

Kovac’s reagent is a biochemical reagent consisting of alcohol (1-pentanol), para-dimethylaminobenzaldehyde (DMAB) and hydrochloric acid. It is used for the diagnostic indole test. The indole produced yields a red complex with DMAB under the given conditions. Red coloration develops when the microorganisms possess tryptophanase activity. A negative reaction shows no color change.
