Apo A1 (Apolipoprotein A1) is synthesized in the liver and small intestine, and is regulated by the dietary intake of lipids. Apo A1 is the major constituent of human high density lipoproteins (HDL) and plays an important role in metabolic and transport function of lipoproteins of the serum. The molecular mass of Apo A1 is 24 kDa and the plasma concentration is range is 270-380 mg/100 ml. Apo A1 is regulated by the dietary intake of lipids, and acts as an activator for the enzyme lecithin cholesterol acyl transferase which converts cholesterol to cholesteryl esters. During lypolysis in the peripheral tissue, Apo A1 can also play a role as an acceptor for surface components of very low density lipoproteins and chylomicrons. The gene encoding Apo A1 is found on chromosome 11. Apo A1 is a cofactor for lecithin cholesterolacyltransferase (LCAT), the enzyme responsible for the formation of most plasma cholesteryl esters. Defects in ApoA1 are associated with HDL deficiency, Tangier disease, and systemic non-neuropathic amyloidosis. At least 9 distinct polymorphic forms of apolipoproteins are known. The apolipoproteins act as stabilizers of the intact lipoprotein particles. Quantitative measurements of HDL, LDL (low density lipoproteins) and VLDL (very low density lipoproteins) particles in human serum are often used to estimate an individuals′ relative risk of coronary heart disease. In addition, quantitative immunological measurements of certain apolipoproteins (especially A-1 and B) have been suggested to be more accurate estimators of coronary heart disease than measurements of lipoprotein particles (especially HDL and LDL). The apolipoproteins are derived from human plasma by density gradient ultracentrifugation and HPLC.Specifications
