VEGFR2 (Vascular Endothelial Growth Factor Receptor 2) is a critical gene located on chromosome 4q12, encoding a receptor tyrosine kinase involved in vascular development and angiogenesis. The receptor structure encompasses seven immunoglobulin-like domains in the extracellular region, a single transmembrane helix, and an intracellular tyrosine kinase domain. Upon binding to VEGF ligands, VEGFR2 undergoes dimerization and autophosphorylation, activating downstream signaling pathways such as the PI3K-Akt, MAPK, and PLC gamma cascades. These pathways regulate endothelial cell proliferation, migration, survival, and permeability, playing a vital role in physiological processes like wound healing and pathological conditions such as cancer and diabetic retinopathy.
