PTP1B (Protein Tyrosine Phosphatase 1B), encoded by the PTPN1 gene located on chromosome 20q13.1, is a major regulator of several cellular signaling pathways, impacting diseases like diabetes, obesity, and various cancers. The protein structure includes multiple exons and functions as a non-transmembrane enzyme that negatively regulates signaling cascades, including insulin and leptin signaling, which are vital for metabolic homeostasis. PTPN2 (Protein Tyrosine Phosphatase Non-Receptor Type 2), located on chromosome 18p11.3-p11.2, also known as T-cell PTP, is similarly a non-transmembrane protein tyrosine phosphatase that modulates immune responses and cell growth. It has ten exons and produces two splice variants, impacting signaling pathways by regulating the phosphorylation state of key proteins.
